Without knowing more about your cycles, it's hard to say. It's not a contraindication. But, we do tell our patients that during the cycle, your pain receptors are on high alert, and botox injections do hurt more during your cycle. Also if you have any issues with temp regulation, like you tend to sweat more or have night sweats, you might sweat out the botox which lessen the duration or how long your botox will last.

If this lesion was 100 % benign, I would take it off using liquid nitrogen. This is the cheapest way to get rid of this mole.
However, if you state this is growing and changing and concern for skin cancer, then I would do excisional biopsy to remove the whole lesions and send the tissue in for pathology.

I believe you are having more anxiety after your first experience with fillers. I would suggest trying melatonin at night to help sleep. I expect the anxiety to go down after couple of weeks of looking good and not having any other adverse side effects from injecting this area.

Here is a partial list of what you can do to increase collagen production in your skin.
fillers - Sculptra or Bellafill are collagen stimulating fillers you can try.
microneeding - this is the same as you trying to needle your skin to stimulate collagen growth. The normal saline is not going to stimulate collagen. Your body is full of normal saline already and you don't see lot of collagen growth everywhere.

If you did not put any fillers in your nose, the changes that are happening to your nose and rest of your face is due to natural process of aging. If you did put filler in the nose, there is a chance you may have developed a scar tissue from the injections.
